    应用3H-TdR掺入实验,RNA印迹分析和反转录-多聚酶链反应等方法观察碱性纤维母细胞生长因子对大鼠血管平滑肌细胞DNA合成及细胞增殖相关基因表达的影响。结果发现,碱性纤维母细胞生长因子作用于血管平滑肌细胞6h后,3H-TdR掺入开始增加,24h达到高峰;在一定浓度范围内,碱性纤维母细胞生长因子对血管平滑肌细胞的促增殖效应与浓度呈正相关。碱性纤维母细胞生长因子作用于血管平滑肌细胞1h后C-fos和c-myc表达活性达到高峰,增殖细胞核抗原在一个细胞周期中出现两次表达。本文结果提示,碱性纤维母细胞生长因子可显著促进血管平滑肌细胞的DNA合成,证实其作用是通过诱导与细胞增殖有关的基因进行表达来实现的。

    Abstract:

    Aim To investigate the effects of basic fibroblast growth factor(bFGF)on the DNA synthesis of vascular smooth muscle cells(VSMC)and the expression of genes related to cell proliferation.Methods The effects of basic fibroblast growth factor on the DNA synthesis was studied by means of 3H-TdR incorporation c-fos,c-myc and proliferating cell nuclear antigen (PCNA) mRNA were displayed by Northern blot and reverse transcription polymerase chain reaction (RT-PCR) respectively.Results The results indicated that R-TdR incorporation into VSMC began increased in 6 h after addition of bFGF, and reached a peak in 24 h. VSMC DNA synthesis was induced by bFGF in a dose dependent manner in certain range of concentrations. The maximal expression of proto-oncogenes c-fos and c-myc mRNA was observed at 1 h after addition of the cytokine.PCNA mRNA expression occured twice in a cell cycle.Conclusions our findings demonstrated that bFGF may significantly stimulate DNA synthesis of VSMC,and suggested that increase in VSMC DNA synthesis was followed by the expression of proto-oncogenes c-fos and c-myc,which is believed as key step in cell proliferating.
