摘要:

目的探讨洛伐他汀对人脐静脉内皮细胞表达血凝素样氧化型低密度脂蛋白受体mRNA和摄取氧化型低密度脂蛋白的影响。方法用TRIzol试剂常规方法提取人脐静脉内皮细胞总RNA,逆转录聚合酶链反应检测血凝素样氧化型低密度脂蛋白受体mRNA含量,检测125I标记的氧化型低密度脂蛋白放射活性法来确定内皮细胞摄取率,分光光度法测量乳酸脱氢酶含量。结果人脐静脉内皮细胞与1、10和50μmol/L洛伐他汀孵育48h后,血凝素样氧化型低密度脂蛋白受体mRNA的表达分别为对照组的84%、69%和48%;与10μmol/L洛伐他汀孵育24、48和96h后,血凝素样氧化型低密度脂蛋白受体mRNA的表达分别为对照组的82%、69%和52%。洛伐他汀使人脐静脉内皮细胞对氧化型低密度脂蛋白的摄取减少,对乳酸脱氢酶含量没有影响。结论洛伐他汀能降低血凝素样氧化型低密度脂蛋白mRNA的表达和对氧化型低密度脂蛋白的摄取。

Abstract:

Aim Toinvestigatewhetherstatinsaltertheexpressionoflectin likeoxidizedlowdensitylipoproteinreceptor (LOX) 1mRNA anduptakeofoxidizedlowdensitylipoproteinonculturedhumanumbilicalveinendothelialcells (hUVEC) . Methods TotalRNAwasextractedwithTrizolreagent. Reversetranscriptionpolymerasechainreaction (RT PCR)wasusedto quantifymRNAexpressionofLOX 1inhUVEC . Ox LDLwasradioiodinatedwith12 5 Itodetecttheuptakeofox LDLbyendothe lialcells. AspectrophotometricenzymeassaywasperformedtodeterminetheactivityofLDH . Results LOX 1mRNAexpressionwassignificantlysuppressedto 84 % ,6 9%and 4 8%ofcontrolafterhUVECwereincubated with 1,10 ,5 0 μmol/Loflovastatinfor4 8h. LOX 1mRNAexpressionwassignificantlysuppressedto 82 % ,6 9%and 5 2 %of controlinthecellstreatedwithlovastatinafter2 4 ,4 8or 96hrespectively . Conclusion LovastatinreducestheexpressionofLOX 1mRNAanduptakeofox LDLwithoutanytoxiceffectswhichmaycon tributetotheantiatheroscleroticpotentialoflovastatin .
