Adipophilin Facilitate Acyl-Coenzyme A∶Cholesterol Acyltransferase 1 Expression in RAW264.7 Cells

    Abstract:

    Aim To explore whether adipophilin affects the expression of acyl-coenzyme A∶cholesterol acyltransferase 1(ACAT1) and illuminate the lipid-accumulation mechanism mediated by adipophilin. Methods The recombinant retroviral vector pQCXIP-HA-Adi was constructed and transfected into packaging cell PA317 mediated by SofastTM,which can induce retrovirus release.Then this retrovirus were used to infect RAW264.7 cell and got RAW264.7 cell line which highly expressed adipophilin.The mRNA level of adipohilin and ACAT1 was determined by reverse transcription-polymerase chain reaction(RT-PCR).The adipophilin and ACAT1 proteins were analyzed by Western blotting.After that,the cells were incubated with atorvastatin for investigating the expression of ACAT1 again. Results After infecting with the retrovirus,both adipophilin mRNA and protein up-regulated,so did ACAT1. However,no effect on ACAT1 expression was found in blank virus transfected cells.The expression of ACAT1 still increased in high-adipophilin-expression cells after using atorvastatin to inhibit endogenous cholesterol synthesis and starving to inhibit exogenous cholesterol. Conclusion Overexpressed adipophilin upregulated ACAT1 expression in RAW264.7 cells.
