Effect of TSG on Expression of E-Selectin, P-Selectin in Vascular Endothelial Cell Induced by H2O2

    Abstract:

    AimTo study the effect of 2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucoside (TSG) on expression of adhesion molecules P-selectin, E-selectin induced by H2O2 in human umbilical vein endothelial cells.MethodsThe expression of P-selectin, E-selectin mRNA and protein were detected by reverse transcription polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA) respectively.Results Treating endothelial cells with 200 μmol/L H2O2, the expression of E-selectin, P-selectin mRNA and protein level were significantly higher (p<0.01).Pretreated with TSG for 4 h, the expression of P-selectin, E-selectin mRNA and protein induced by H2O2 were significantly inhibited in endothelial cells (p<0.05).ConclusionsTSG probably can protect human umbilical vein endothelial cells from oxidative stress by decreasing expression of adhesion molecules.
