    • Calcium Dependency of Monocyte Migration Induced by Smooth Muscle Cell-derived Chemotactic Factor

      Abstract:The monocyte migration induced by the media, as the source of chemotactic factor,conditioned by cultured rabbit aortic smooth muscle cells, was assayed by micropore filter method using modified Boyden chamber;the effects of extracellular Ca2+ at different concentrations and verapamil on the monocyte migration were observed as well.Furthermore, the influences of above-mentioned conditioned media and verapamil on monocyte cytosolic free Ca2+ concentration, [Ca2+]i, which was determined by using Fura-2 measurement, were observed. The results showed that the conditioned media were significantly chemotactic for monocytes and its chemotactic activity was inhibited by verapamil; the media also significantly increased [Ca2+]i that were inhibited by verapamil as well.Extracellular Ca2+ had also some influences on monocyte migration. The results suggest that monocyte migration are dependent on both cytosolic free Ca2+ and extracellular Ca2+.

    • LDL Receptors of Lymphocytes and Binding of LDL by Non-parenchymal Cells of Rat Liver in Endogenous Hypertriglyceridemics

      Abstract:A study on LDL receptors of lymphocytes and plasma LDL binding activity to liver non-parenchymal cells of rat in 5 normal persons and endogenous hypertriglyceridemlcs(whose plasma triglycerides levels were 0. 87±0. 32 and 3.07 ±1.67 mmol/L respectively) was made. Plasma LDL was isolated by density gradient ultracentrifugation. LDL receptors of lymphocytes were measured by a enzyme-linked method developed by this laboratory.125 I-labelled-LDL was prepared by IC1 method. Liver non-parenchymal cells(NPC) of rat were prepared by collagenase method.125ILDL binding to rat liver NPC was measured by Goldstein and Brown' s method. The results showed that the Bmax of LDL binding to lymphocytes from hypertriglyceridemic group was significantly higher than that of normal group(57.5±13.2:38.2±9.3 ng/mg cell protein,p<0. 05), but there was no difference in affinity(Kd) between the normal and hypertriglyceridemic groups(12.0±3.2:13.3±4.1μg/ml, p>0. 05).The binding activity of plasma LDL from hypertriglyceridemics to rat liver NPC was significantly lower than that of LDL from the normal group(p<0. 05). The results are disscused.

    • Effect of Hormone-sensitive Lipases Overexpression on the Formation of Chinese Hamster Ovary Cell Derived Foam Cell

      Abstract:A foam cell model was developed by exposure of Chinese Hamster ovary cells to 20μg/ml β-VLDL. In this foam cell model hormone-sensitive lipase(HSL) activity was overexpressed using adenovirus complexed with the rat HSL gene.Nearly 300 folds increase of intracellular cholesteryl ester hydrolase activity was achieved in transfected cells compared with the control cells. When cells overexpressing HSL were incubated with β-VLDL,cholesteryl ester levels were only 25% of those seen in control cells. Thus, HSL strongly inhibits cholesteryl ester accumulation in this foam cell model.

    • Preparation and Characterization of Monoclonal Antibodies Against Low Density Lipoprotein Receptor of Rabbit

      Abstract:In this study,the partially purified low density lipoprotein(LDL) receptor from rabbit liver was used as an antigen, and two monoclonal antibodies were obtained, designated as ZMRE6 and ZMRF7 respectively, both of them can react specifically to rabbit LDL receptor with a high titer and belong to the subgroup of IgG1.Tested with indirect immunofluroscent staining, no cross reaction was found between these two monoclonal antibodies and the cultured human fibroblasts and rhesus endothelial cells,so it is considered that ZMRE6 andZMRF7 have a genus-specificity.

    • Expression of Endothelin and Nitric Oxide synthetase in Rabbit's Atherosclerotic Plaques

      Abstract:Present test observated the synthesis and expression of endothelin(ET) and nitric oxide synthetase(NOS) in rabbit' s atherosclerotic plaque(AP) on polypeptide and gene transfer.Histochemistry and immunohistochemistry shown that in proliferating vascular smooth muscle cell (VSMC) within AP, synthesis of ET was increased while that of NOS was significantly decreased;Meanwhile, in situ hybridization also shown that the transfer of ET mRNA was great increased while that of NOS mRNA was great decreased within 5 cases and only within 1 case,a little of NOS mRNA was expressed and the range was limited.So we believed that in aortic VSMC, un-balence of between ET and NOS may be involved in the occurrence and development of AP.

    • c-myc Expression and Vascular Smooth Muscle Cell Proliferation After Rat Aortic Injury

      Abstract:To understand the molecular events governing smooth muscle cell(SMC) prolifetation in vivo, SMC proliferation and c-myc expression were assessed in aorta in 25 male rats. Balloon-injury to rat aorta evoked platelet adhesion after 24 h and induced increases of SMC proliferation and intimal thickening after 5, 10 and 15 days. c-myc expression increased significantly 24 h, but decreased gradually 5, 10, 15 and 20 days after the injuryThe results indicate that c-myc gene expression corroborates the platelet adhesion and autocrine mode of smooth muscle cells.

    • The Metabolism of Low Density Lipoprotein Isolated from Diabetic Patients in Human Monocyte-derived Macrophages

      Abstract:Macrophages are proved to be the precursors of the lipidladen foam cells that characterize early atherosclerotic lesion. Hyperglycemia in diabetic patients lead to the high concentration of glycated low density lipoprotein(LDL) and abnormal metabolism of LDL. In this study, we used radioreceptor assay to compare the metabotic results of LDL from diabetic patients and from nondiabetic control subjects. We found that both uptake and degradation of diabetic LDL were significantly higher than that of control LDL. We also discovered that the total cholerslerol content of cultured macrophages incubated with diabetic LDL significantly increased as compared with that incubated with control LDL. This study suggests that interaction between glycated LDL and macrophages accelerates formation of foam cells, and contributes to the accelerated atherosclerosis of diabetes.

    • The Increase of c-myc and hsp70 Gene Expression in Rat Aortae Induced by Perfusing Endothelin in vitro

      Abstract:In the study rat aortae were perfused by endothelin in vitro and the RNA samplewas extracted from them for Northern blotting in which c-myc and hsp70 gene probes were labeled by α-32P-dCTP,The results on the bands of autoradiographs show that c-myc and hsp70 gene expression increases in the rat aortae. It might play an important role on proliferation of vascular smooth muscle cells in hypertension and atherosclerosis.

    • Early Changes of Coronary Atherosclerosis in Fetuses at Different Ages

      Abstract:We collected more than 400 fetuses at different ages. out of them 120 cases were taken randomly whose ages ranged from 3 to 9 months. They were divided into 8 groups according to the age. Sections were made from the left and right coronary arteries where atherosclerosis occurs most commonly.All sections were stained with hematoxylin-eosin, Weigert-VG mixture,Masson' s trichrome and Alcian Blue-PAS,In addition, some of them were stained with fat and mucins.The results were as follows:1.Early changes of coronary atherosclerosis of the foetuses at different ages.①Fat spots and fatty streaks. We believe that they can develop into atherosclerosis or stop developing or subside,②Proliferation focuses of smooth muscle cells.Smooth muscle cells are the main elements of pathological changes of atherosclerosis, participating in the formation of atherosclerosis.③Smooth muscle cells can produce fibrous connective tissues and mucopolysac charide in the walls of coronary arteries. The above mentionded findings in the fetuses are of great significance in exploring the occurrence and development of atherosclerosis.2.1~Ⅱ grade positive pathological changes of coronary atherosclerosis were found in the fetuses.It protruded from the media through the intima into the vascular cavities, occupying 255 or more of the cavity.It had made the vascular cavities narrow to a degree of 1~Ⅱ grade. It is not easy to subsidecompletely. The living conditions and environments of the fetuses are simpler than that after their birth, and these are of specific importance in studying the etiology and pathogenesis of atherosclerosis.Since the nourishment of the fetuses are supplied by the blood of the mothers' uteri, whether the early pathological changes in the coronary artery and Ⅰ ~Ⅱ grade positive pathological changes of the fetuses are due to the hyperlipidemia of their mothers or the hereditary factors needs to be studied.

    • Plasma Lipids of Type Ⅱ Diabetics Before and After Zincic-tonic Treatment and Its Clinical Significance

      Abstract:The plasma lipids of 44 type Ⅱ diabetics and 30 healthy persons were compared and analysed, the diabetics were divided into two groups. Group A(n=225) treated with D860 and glucozinc, while Group B only with D860, the course of treatment of both group was one month.The plasma lipids of the diabetics in both group and the healthy persons were measured before and after the treatment respectively. The results showed that:① The triglyceride(TG)and the low-density lipoprotein cholesterol(LDL-C) of the diabetics in both group were distinctly higher than that of the healthy persons, while the high-density lipoprotein cholesterol(HDL-C) was lower than that of healthy controls, ② After the treatment, the HDL-C of Group A was remarkably raised while the TG and LDL-C remarkably reduced, as for Group B, no distinct change was found in HDL-C. This study suggests that Zincic-tonic has an effect to raise the HDL-C level of diabetics who are, at the sametime,in treatment with plasma glucose-lowering medicines.
