Abstract:The cultured porcin artery smooth muscle cells became foam cells by preeincubated with 10 mg·L-1 oxidized low density lipoprotein for 72 hours, which could accumulate cholesteryl ester and the ratio between cholesteryl ester and total cholesterol was 64. 1%. When the foam cells were incubared with 0. 01 mg·L-1 and 0. 1mg·L-1 Xiaoban peptide for 24 hours, they reversed to normal smooth muscle cells, whose ratio between cholesteryl ester and total choles terol was less than 50%. The amount of nitric oxide increased in the medium of foam cell and decreased in the proess of reversing to normal level. It suggest that Xiaoban peptide may be a new antiatherosclerosis medicine.

Abstract:In this study, a typical model of macrophages-derived foam cell was founded by peritoneal macrophage of C57 BL/6J mouse, which is susceptible to atherosclerosis, incubated with 10 mg·L-1 oxidized low density lipoprotein. Comparing with macrophages of Swiss strain mouse, these results showed the cultured foam cell-derived from C57 BL/6J mouse macrophages continued a large of lipid particles and intracellular total cholesterol increased in the cells, in which cholesteryl ester occupied more than 50% of total cholesterol. It corresponded to the typicalcharacteristics of foam cells. This model suppled a reliable pathological cell tool for the study of atherosclerosis.

Abstract:Total RNA of normal human aortic smooth muscle and fatty streaks from thoracic aortawas isolated by acid guanidium thiocyanate-phenolchloroform extraction. The gene expression of PDGFA, B chain and PDGF α- and β-receptor was investigated by Northern bolt analysis. Results In normal aortic smooth muscle or the normal region from the aorta with atherosclerotic lesions, the gene expression of PDGF was very low, while that of PDGF receptor was not detected. In contrast, in the fatty streaks, the expression of PDGF A mRNA and PDGF β-receptor mRNA were augmented significantly. Conclusions It suggests that the increased gene expression of PDGF A chain and PDGF β-receptor pro motes the proliferation of smooth muscle cells thus highly related to the pathogenesis of atherosclerosis.

Abstract:With mimicking the injury model from murine peritoneal macrophages attacked by oxidized low density lipoprotein (OLDL), the time-de pendant and concentration-depandant effect of polysac charide K (PSK) on the foam-like change and attenuation of murine peritoneal macrophages attacked by OLDL was observed.After incubated with 10,50 and 100 mg·L-1 OLDL for 24,48,72 hOurs in vitro, the fingure of PSK-treated macrophages was maintained normal relatively.Moreover,in PSK-treated group, the amount of survival cells were higher with counted by MTT colorimetric analysis at every time points.So PSK could inhibit the foam-like change of macrophages efficiently.And after the change in macrophages formed,PSK could prolong the survival time of these macrophages.

Abstract:To purify a smooth muscle cell derived growth factor,the serum free medium conditioned by cultured rabbit aortic smooth muscle cells(SMC-CM)was collected.Methods The SMC-CM was 5.8 fold concentrated by ultrafiltration using Millipore Ultrafiltration System with a molecular weight cut off at 10 kDa,and then was purified by heparin affinity chromatogrphy using a column of heparin-Sepharose CL-6B.Incorporation of3H-thymidine(3H-TdR)into cell DNA was used to measure the mitogenic activity of the fractions from chromatography for NIH 3T3 fibroblasts. The molecular weight and the iso-electric point of these fractions were determined by NaDodSO4-polyacrylamide gel electrophoresis (SDS-PAGE) and iso-electric focusing, respectively. Results The protein eluted in 1.0~1.6 mol·L-1 NaCl from the heparin-sepharose was mitogenic for 3T3 cells, and this protein had a molecular weight of 23.0~26.9 kDa and iso-electric point of about 4.6. Conclusions The fact that the above-mentioned biochemical properties differed from that of PDGF, IGF and FGF suggests that this mitogenic protein may be a separate growth factor.

Abstract:It has been recognized for many years that adenosine and adenosine 5'-monophosphat (AMP),adenosine 5'-diphosphate(ADP)and adenosine 5'-triphosphat(ATP)are potent vasoactive substances.In this paper,it was demonstrated that ATP,ADP induced endothelium dependent relaxation on isolated porcine coronary arterial rings but adenosine produced endothelium-independent relexation on aboveprepations.It was also observed with MTT colorimetric assay that AMP,ADP and ATP inhibited proliferetion of porcine aortic smooth muscles in dose-dependent manner, similar to adenosine. The results suggest that purine nucleoside adenoside and its analogues act as growth regulators of vascular smooth muscle cells. In addition,the clinical application of adenosine drugs was discussed in detail.

Results Cultured rabbit peritoneal exudate macrophages produced a factor that was chemotactic for monocytes,and the activity due to chemotaxis and not chemokinesis,the chemotactic activity was markedly inhibited by anti-MCP-1 antibody. MCP-1 mRNA was expressed by the macrophages.Conclusions Macrophages can produce MCP-1 and play an important role in atherogenesis through attracting monocytes to migrate into subendothelial space.

Abstract:用免疫组织化学方法研究北京、南宁和宁波青年人冠状动脉内膜平滑肌细胞中结蛋白和前胸腺素的表达情况。测得结蛋白阳性率的高低顺序为南宁、宁波、北京。南宁与北京的差别有显著性(P=0.0004),宁波与北京的差别有意义(P=0.019),而南宁与宁波的差别无意义(P=0.2156)。结蛋白可作为肌细胞分化的标志,提示南宁与宁波青年冠状动脉内膜平滑肌细胞向成熟分化,而北京人则缺乏这种倾向。前胸腺素阳性率顺序为北京、南宁、宁波。提示北京人冠状动脉内膜SMC相对有较高的增长趋势。上述结果显示高低发区SMC内部结构的差别。

Abstract:分析1985~1991年老年组106例及非老年组116例心肌梗塞的机械并发症。老年组乳头肌功能失调占37.7%,高于非老年组的17.2%(p<0.01);室壁瘤(VA)72.1%,高于非老年组的55.6%(p<0.01);二组心脏破裂(CR)发生率差异无显著性。分析老年组心肌梗塞的机械并发症的影响因素:无Q波心肌梗塞乳头肌功能失调占73.7%,大于有Q波心肌梗塞中所占29.9%(p<0.01)。有Q波心肌梗塞中,左室内径扩大者乳头肌功能失调占56.1%,高于左室内径正常者的20.0%(p<0.01)。左室内径扩大及乳头肌功能失调导致左室射血分数明显下降。心室壁增厚、无心绞痛病史及首次发生的有Q波型心肌梗塞易导致室壁瘤形成。在广泛前壁心肌梗塞患者易发生心脏破裂。

Abstract:Serum lipids, apolipoprotein C(apo C)levels and post-heparin lipoprotein lipase (LPL)and hepatic triglyceride lipase (HTGL)activities were measured in 30 patients with coronary heart disease (CHD),20 hypertriglyceridemic subjects and 20 control subjects.The results showed that the level of serum apo C Ⅱ in CHD1 patients,with serum triglyceride (TG)<1.41 mmol·L-1,had siginficantly lower level compared to control group, which were reduced 14. 8%, while level of serum apo CⅡ in CHD3 patients,with serum TG > 2. 03 mmol·L-1, were increased significantly.The level of serum apo CⅡ in hypertriglyceridemic patients had significantly higher than that in control group and revealed positive correlation with serum TG (r=0. 599, p<0. 01). Compared with controls, LPL activities in CHD groups were decreased markedly (p<0. 001). This indicates that it may be important significance clinically to measure serum apo C Ⅱ levels for CHD1 group. Increasing apo C Ⅲ levels of hypertriglyceridemic subjects were closely related to the elevated TG levels, the elevation of apo C Ⅲ levels was likely an important cause of forming hypertriglyceridemia. Low levels markedly of LPL activity in CHD patients may be the causes of TG elevated and high density lipoprotein (HDL)decreased,measuring LPL activity may be helpful for diagnosing CHD clinically.

Abstract:On the basis of observation that nitrendipine has a role of anti-atherosclerosis,the effects of nitrendipine on experimental atherosclerosis of rabbits was studied. Results showed that nitrendipine might inhibit the increase of platelet aggregation and lipid peroxidation induced by hypercholesterolemia,which suggest that the effect of nitrendipine antiatherosclerosis is linked with thses changes.

Abstract:The paper reported the effect of fibrinolysin from Hunan Agkistrodon acutus on hemorheology in 23 cases of hyperviscosaemia and 31 cases of cerebral thrombosis. It is found that the flbrinolysin from Hunan Agkistrodon acutus may reduce low shear of blood specific viscosity, erythrocyte sedimentation rate (ESR),equation K value of ESR,and aggregative index of red blood cell, fibrinogen and plasma specific viscosity in patients with hyperviscosaemia and improve microcirculation of hyponychium in patients with cerebral thrombosis.This results indicate that the fibrinolysin may be used to treat these diseases.

Abstract:Apolipoprotein CⅠ,CⅢwere isolated and purified by ultracentrifugation, gel filtration and high performance liquide chromatography from human serum very low density lipoprotein. The purity of apo CⅠ and CⅢ was characterized by IEF、SDS-PAGE and double immunodiffusion. The BALB/C mice were immunized by purified apo CⅠ or CⅢ.4 lines of hybridoma to apo CⅠ and 2 lines of to apo CⅢwhich were able to secreat monoclonal antibOdies(McAb) to human apo CⅠ and apo CⅡ respectively were established. The McAbs have no cross reaction to apo AⅠ,AⅡ,B,CⅡ,E and human albumin.
