Abstract:Aim To investigate the regulation of X-box binding protein-1 (XBP-1) and acyl-CoA cholesterol acyltransferase-2 (ACAT-2) expression by cholesterol in hepatic cell and enterocyte. Methods HepG2 cell and Caco2 cell were incubated with different concentration of free cholesterol and oxidative cholesterol. The mRNA level of XBP-1 and ACAT-2 in cells were detected by semi-quantitative RT-PCR. Results In cells incubated with free cholesterol and oxidative cholesterol, XBP-1 and ACAT-2 mRNA levels in HepG2 cell and Caco2 cell were up-regulated, and were dependent on the concentration increase. Conclusions Both of cholesterol and oxidative cholesterol can induce UPR, and ACAT-2 mRNA level can be up-regulated, the results imply the possibility that ACAT-2 may be regulated by UPR; in view of ACAT-2 playing an important role in the cholesterol esterification and absorption, UPR may have important significance in cholesterol absorption.

Abstract:Aim To study the relationship between serum bilirubin levels and coronary artery disease. Methods 120 patients undergoing diagnostic coronary arteriography(CAG) were divided into two groups:the control group(the results of CAG is normal) with 40 patients, the coronary heart disease group(≥50% diameter narrowing) with 80 patients.15 cases of coronary heart disease group were mild,32 cases of coronary heart disease group were moderate and 33 cases of coronary heart disease group were severe. The serum bilirubin was determined with Malloy-Evelyn.Results Serum Level of bilirubin in coronary heart disease group were statistical significantly compared with control group(p<0.05).There was positive correlation between the serum level of bilirubin and the degree of coronary aretry narrowing(r=-0.26,p<0.05).Conclusions Low serum level of bilirubin may be a new dangerous factor for coronary heart disease. There is correlation between serum level of bilirubin and the degree of coronary aretry narrowing.

Abstract:Aim To investigate the changes and the clinical significances of blood lipids in patients with systemic lupus erythematosus(SLE).Methods Retrospective analysis was done on the changes of blood lipids in 85 SLE patients who hadn't been treated with steroid,56 SLE patients who had been treated with steroid and 60 healthy individials. Results The level of triglyceride(TG),total cholesterol(TC) and low density lipoprotein cholesterol(LDLC) were higher in SLE patients who had been treated with steroid than that of control and that of SLE patients who hadn't been treated with steroid(p<0.05),but had no significant difference between SLE patients who hadn't been treated with steroid and control(p>0.05).The level of blood lipids was abnormal in 51.8%(44/85) of the patients who hadn't been treated with steroid, and in 22/25 patients who had renal involvement. It had significant difference between SLE patients who had renal involvement and control(p<0.01).Conclusions The levels of blood lipids were abnormal in SLE patients. It is associated with renal involvement and high dose corticosteroids. The measurement of blood lipids is important for the patient with SLE to prevent and treat the complication of the disease of high blood lipids.
