Abstract:AimTo investigate the effects of Jagged1 over-expression on aged rat-derived endothelial progenitor cells migration and proliferation.MethodsMononuclear cells were obtained from bone marrow of young (1 to 2 month-old) and aged (19 to 26 month-old) Sprague-Dawley rats by Ficoll density gradient centrifugation and cultured with medium DMEM/F12, 48 h later, the suspending cells were translocated to be cultured in new flasks coated with fibronectin, the secondary attached cells were used to perform the further experiments.EPCs were characterized as double positive for Dil-ac-LDL uptake and lectin binding and cells were further identified by vWF expression.The experiments were divided into four groups: control group (aged rat-derived EPCs without gene transfection), PIRES2-EGFP transfection group, PIRES2-EGFP-Jagged1 transfection group and young rat-derived EPCs group in which transfection was also not performed.The GFP expression cell number was acquired through fluorescence microscope and transfection efficiency was calculated.Immunofluorescence, RT-PCR and Western blot were used to detect the mRNA and protein expression respectively.Cell migration and proliferation were tested by Transwell culture and MTT assay respectively.ResultsImmunofluorescence showed that gene transfection markedly promoted Jagged1 expression; RT-PCR and Western blot demonstrated that Jagged1 mRNA and protein expression significantly increased in aged rat-derived EPCs (P<0.01) compared

Abstract:AimTo study the effect and mechanism of rhynchophylla total alkaloids, sinapine cyanide sulfonate and its component compatibility on change of vascular endothelial cells induced by the tumor necrosis factor-α(TNF-α).MethodsRat vascular endothelial cells were cultured in vitro, and vascular endothelial cell injury model was established by TNF-α.Then changes of cell morphology were observed before and after treatment; intercellular adhesion molecule-1(ICAM-1), vascular cell adhesion molecule-1(VCAM-1) and rat endothelin-1 (ET-1) were determined by Elisa method, mRNA of ICAM-1, VCAM-1 and ET-1 by RT-PCR method and the level change of endothelium-derived relaxing factor (NO) by nitrate reductase method.ResultsThe vascular endothelial cell injury model could be established in vitro successfully by TNF-α (5 μg/L).Compared with TNF-α group, treated groups could improve vascular endothelial cell morphology, lower the secretion of ICAM-1,VCAM-1 and ET-1.Compared with rhynchophylla total alkaloids group and sinapine cyanide sulfonate group, the component compatibility group was better on heightening the secretion of NO, down-regulating the expression of ICAM-1 mRNA(P<0.05).On down-regulating the expression of mRNA of VCAM-1

Abstract:AimTo study on changes of infarcted border zone of effective refractory period ( ERP ), and expression of connexin43(Cx43) , Tyrosine hydroxylase(TH)and nerve growth factor (GAP43 ) in MI SD rat by echocardiograph,electrophysiological techniques and immunohistochemical technique.To explore relation of left ventricular remodeling, electrical remodeling and sympathetic nerve remodeling of myocardial infarction rat.Methods30 SD rats of either sex weighting 220~250 g were euthanized under chlorine hydrate 0.3 mL/100 mg anesthesia and were randomly divided into two groups: sham operation group (n=15), MI group(n=15).ResultsCompared with the sham operation group, LVIDd、LVIDs were significantly increased and EF, FS were shortened in MI group (P<0.01).Compared with the sham operation group, ERP of infarcted border zone was significantly shortened in MI group(P<0.01).Left ventricular CX43 immunofluorescence could be seen under the fluorescence microscope in two groups of rats.the fluorescence became weak in MI group.Immunohistochemistry of left ventricular 43 and TH could be seen under the light microscope in two groups of rats.Compared with the sham operation group, GAP43 and TH had significantly higher expression in MI group (P<0.01), respectively.ConclusionsVentricular remodeling, sympathetic remodeling and electrical remod-

Abstract:AimTo clarify the effects of myocardial apoptosis on ventricular remodeling after myocardial infarction, and to study the effects of mast cell degranulation on myocardial apoptosis and fibrosis.MethodsMyocardial infarction (MI) model was established in male Sprague-Dawley (SD) rats by ligation of left anterior descending coronary artery.The rats were randomly divided into three groups: sham-operated group (n=6), MI model group (n=8) and cromolyn sodium group (n=7).Four weeks after ligation, myocardial apoptosis index (AI), collagen volume fraction (CVF) and expression level of caspase-3, Bcl-2 and Bax were obtained.ResultsCompared with sham-operated group, AI, CVF and expression level of caspase-3 and Bax in MI model group and cromolyn sodium group increased (P<0.05), but decreased in Bcl-2 (P<0.05).Cromolyn sodium group was decreased in these factors (P<0.05) compared with MI model group.Bcl-2 level in cromolyn sodium group was higher than MI model group, but there is no significant difference among these two groups (P=0.127).The AI was positive correlation with CVF (r=0.769, P<0.01).
Abstract:AimTo investigate the effect of low-dose aspirin on kidney in diabetic rats and discuss possible mechanism.MethodsMale SD rats were injected streptozotocin (STZ) 30 mg/kg by tail intravenous for establishing the type 1 diabetes animal model, which were randomly divided into diabetes model group and 9 mg/kg aspirin i.e. acetylsalicylic acid (ASA) treatment group, and other 8 healthy male SD rats were included into the control group.The rats took medicine by irrigation stomach for 12 weeks, then their fasting blood glucose, blood urea and serum creatinine, and urine protein content were inspected, kidney tissue pathology microstructure was inspected by HE dyeing method, and Caspase-3, Bax and Bcl-2 expression was detected by immunohistochemical method.ResultsCompared with the normal control group rats, in diabetes model rats, the content of fasting blood glucose, blood urea,serum creatinine and urine protein was increased, glomerular atrophy was more obviously, kidney tissues Caspase-3 and Bax express were enhanced, meanwhile kidney tissues Bcl-2 expression and Bcl-2/Bax ratio were decreased; in ASA treatment rats, the above indexes were improved.ConclusionLow-dose aspirin has protective effect in diabetic rats kidney, which may regulate the expression of cell apoptosis related protein.
Abstract:AimTo investigate the effects of rosuvastatin on neointimal hyperplasia and mitofusin 2 (Mfn2) expression after carotid artery balloon injury in rats.MethodsThirty-two male Wistar rats were randomly divided into sham-operated group, model group, rosuvastatin group 1 and group 2.The rats in the two rosuvastatin groups were given rosuvastatin 5 mg/kg or 20 mg/kg daily, respectively.The rats in the model group and the two rosuvastatin groups were induced neointimal hyperplasia by carotid artery balloon injury.After the rats were sacrificed at day 14, HE staining was used to observe the change of vascular morphology.Immunohistochemistry staining and Western blot were performed to measure the expression of Mfn2 and proliferating cell nuclear antigen (PCNA).Evans blue staining was used to measure the reendothlialized area.ResultsOn the 14 th day after operation, compared with the model group, the neointimal area and intimal/media area ratio were significantly decreased in the two rosuvastatin groups (P<0.01).Mfn2 expression was up-regulated in the two rosuvastatin groups (P<0.01).Rosuvastatin did not inhibit the reendothlialized in the injured carotid arteries.ConclusionsRosuvastatin attenuates neointimal hyperplasia and restenosis in the rat carotid artery balloon injury model, which may be related to up-regulated expression of Mfn2.

Abstract:AimTo research the affect factors and the solutions for diagnosis of coronary stenosis with arteriosclerosis plaques that were divided into two categories (the non-calcified plaques and calcified plaques) with 320-slices computed tomography angiography (CTA) compared with tocoronary angiography (CAG).Methods396 coronary arteriosclerosis plaque lesions were found in 102 patients examed by CTA and CAG.They were divided into five groups according to vascular stenosis degree: normal, mild, moderate, severe and occlusion.The coincidence rate of vascular stenosis degree by CTA were calculated compared with CAG with lesions divided into two categories (the non-calcified plaques and calcified plaques).The calcium threshold for calcified plaques is set in four different programmes: >150 HU, >250 HU, >350 HU, >600 HU.ResultsCTA shows the 396 arteriosclerosis plaques with 68.2% for calcified plaques and 31.8% for non-calcified plaques, and 6% is unstable plaques, 7.8% without vascular stenosis.It is the same as calcium threshold in groups of >150 HU, >250 HU, >350 HU for the coincidence rate of vascular stenosis degree comparing CTA with CAG but lower in group of >600 HU.The coincidence rate comparing CTA with CAG of is

Abstract:AimTo explore the incidence and affecting factors of left ventricular hypertrophy (LVH) in chronic kidney disease (CKD) 3~5 patients.Methods270 CKD 3~5 patients were enrolled from department of nephrology, the 3rd hospital of Sun Yat-Sen University in this study from 2010 to 2012.Ambulatory blood pressure, clinical blood pressure, cardiac ultrasonography, proteinuria, and other clinical data were collected.Results46.67% CKD patients had LVH, in which male accounted for 39.74% and female accounted for 55.46%.Female group had more LVH patients than male group (P<0.05).There were 25.0% CKD 3 patients, 44.9% CKD 4 patients and 53.6% CKD 5 patients with LVH respectively, which had significant difference among the three groups(P<0.05).LVH group had higher intact parathyroid hormone (iPTH), body mass index (BMI), phosphorus, serum creatinine (SCr), left ventricular mass index (LVMI), pulse, PP, left ventricular end diastolic dimension (LVDd), left ventricular posterior wall thickness (LVPWT), interventricular septal thickness (IVST), and higher clinic systolic blood pressure (SBP), ambulatory SBP and diastolic blood pressure (DBP), day and night SBP and DBP(P<0.05), but had lower estimated glomerular filtration rate (eGFR), hemoglobin (HGB), calcium (Ca) and ejection fraction (EF) value(P<0.05) compared with non LVH group.Multivariate linear analysis indicated that BMI, iPTH and night SBP were independently associated with LVMI.

Abstract:AimTo investigate the association of serum high sensitivity C-reactive protein (hs-CRP) and Periostin protein with coronary artery stenosis in coronary heart disease patients.MethodsSerum hs-CRP and Periostin protein were measured using enzyme-linked immunosorbent assay (ELISA) in 87 patients with coronary heart disease, of which 57 patients were diagnosed as acute myocardial infarction (AMI) and 30 were unstable angina pectoris (UAP).Coronary artery stenosis was characterized by Gensini scores.ResultsSerum hs-CRP and Periostin protein were significantly increased in AMI patients compared with UAP (P<0.05 or P<0.001) and they were in positive correlation with Gensini score (P<0.05 or P<0.001).The two protein were also in positive correlation with each other (P<0.05).

Abstract:AimTo evaluate the diagnostic value of 320-detector row dynamic volume CT angiography (CTA) on patients with coronary artery stenosis.MethodsA total of forty consecutive subjects with suspected coronary heart disease were enrolled in this study.CTA and invasive coronary angiography (ICA) were taken consecutively in order to evaluate the diagnostic value of CTA.The sensitivity, specificity, positive and negative predictive value were calculated with the ICA as the gold standard.ResultsNinety-one and 67 lesions with ≥50% luminal stenosis were detected by CTA and ICA respectively.The sensitivity, specificity, positive and negative predictive value of CTA were 93% (95%CI: 84%~97%), 95% (95%CI: 93%~97%), 74% (95%CI: 6%~82%), 99% (95%CI: 98%~100%) respectively, with the ICA as the gold standard.And the area under the curve (AUC) was 0.98 (0.97~0.99).Conclusions320-detector row dynamic volume CT angiography is reliable in diagnosis of suspected coronary heart disease, which provided a non-invasive method for the diagnosis.
Abstract:AimTo explore how to reduce the incidence of coronary artery in-stent restenosis (ISR) via analysis of the related factors to ISR.Methods100 cases with coronary artery ISR and 100 cases without coronary artery ISR were selected.We analyzed the related factors affecting the coronary ISR by comparing the coronary heart disease risk factors and the coronary stent situations between the two groups of patients, and finally obtained the relevant intensity order for related factors by Logistic regression analysis.ResultsSignificant differences (P<0.05) were shown both in smoking, blood pressure, blood sugar, total cholesterol, low density lipoprotein cholesterol, and in the diameter, length, location and quantity of stents between two groups with or without coronary ISR; Among the acute myocardial infarction (AMI) patients, significant differences (P<0.05) were also shown in no application of Ⅱb/Ⅲa receptor antagonist and elevated level of C-reactive protein between two groups.Logistic regression analysis showed that coronary ISR had a strong correlation with 2 h postprandial blood glucose and smoking; The relevant intensity of coronary stents situation to coronary ISR showed: many pieces of stents > stent diameter > stent length > stent location.ConclusionDiabetes, smoking, hypertension, hyperlipidemia, multivessel coronary lesions, severe stenosis, acute myocardial infarction patients without application of platelet Ⅱb/Ⅲa receptor antagonist, elevated level of C-reactive protein are the important factors associated with coronary artery ISR.
Abstract:AimTo investigate the relationship between levels of serum cystatin C (CysC) and carotid athrosclerosis in patients with acute cerebral infarction.MethodsNinety-eight acute cerebral infarction patients treated in our hospital from July 2010 to March 2012 were investigated.All of them were divided into carotid intima-media thickness (IMT) ≥0.9 mm group with 50 patients, and IMT <0.9 mm group with 48 patients, by carotid ultrasonography.Levels of serum CysC and related clinical data were measured.ResultsThe levels of serum CysC in the IMT≥0.9 mm group was significantly higher than those in the IMT <0.9 mm group (0.95±0.24 mg/L vs 0.76±0.15 mg/L,P<0.01).CysC had obvious negative correlation with serum HDLC (P<0.01), while postive correlation with age, body mass index (BMI), systolic blood pressure (SBP), low density lipoprotein cholesterol (LDLC) and IMT (P<0.01).Multinmial regression analysis confirmed that CysC, SBP and LDLC were independent factors correlated with carotid IMT.ConclusionsCysC has associations with carotid atherosclerosis in patients with acute cerebral infarction.CysC could probably be an independent factor correlated with the carotid IMT.
Abstract:AimTo investigate the risk factors of cerebral infarction in cancer patients.Methods67 cases of tumor patients with cerebral infarction were included as the tumor group who were treated in hospital from January 2005 to August 2012, 67 cases of general cerebral infarction were randomly selected at the same times as control group.The difference was analysed between tumor group and control group in hypertension, diabetes, hyperlipidemia,plasma fibrinogen and D- dimmer.ResultsIncidence of hypertension, diabetes and hyperlipidemia was significantly lower in tumor group (P<0.05 or P<0.01), D-dimmer and fibrinogen level was significantly higher than that of contrl group ( P<0.05 or P<0.01).Multivariate regression analysis showed that plasma fibrinogen and D-dimmer were the risk factors of cerebral infarction in cancer patients.ConclusionsStroke risk factors are different between the cancer patients and general people.Cancer-associated hypercoagulation is an important stroke risk factor in patients with cancer.

Abstract:AimTo compare the efficacy of everolimus-eluting versus sirolimus-eluting stents in patients with coronary artery disease.MethodsBy searching Medline(1960~2012.1) , CENTRAL(the Cochrane central register of controlled trials, 1989~2012.1), EMBASE(1980~2012.1), CNKI(1979~2012.1)databases, the open published randomized controlled trials (RCT) about the efficacy of everolimus-eluting versus sirolimus-eluting stents in patients with coronary artery disease was calculated by statistic software Stata11.0.The relative risk (RR) was used as the effect size to compare the events occurred during the follow-up period between the two groups.ResultsSix trials including a total of 5196 patients were analyzed in this study.No statistical difference of major cardiac adverse events (death/ myocardial infarction) (RR=1.00, 95%CI:0.84~1.17,P=0.96), definite stents thrombosis(RR=0.53,95%CI:0.24~1.16,P=0.11) and revascularization (RR=0.88,95%CI:0.70~1.09,P=0.24) were found in this meta-analysis, respectively.ConclusionsThis meta-analysis did not demonstrate significant statistical difference in terms of efficacy and safety between everolimus-eluting and sirolimus-eluting stents in patients with coronary artery disease.

Abstract:AimTo determine the effect of oxLig-1, synthesized by an chemical method, on the monocyte chemoattractant protein-1 (MCP-1) expression in human THP-1 monocytes.MethodsoxLig-1 was identified by mass spectrometry and nuclear magnetic resonance (NMR) analysis.Human THP-1 monocytes were treated with different concentration (15, 30 and 60 mg/L) of oxLig-1, the apoptosis of the cell was detected by flow cytometry using Annexin-V and PI.The mRNA and protein levels of MCP-1 were measured by RT-PCR and ELISA methods, respectively.ResultsThe structure of synthetic oxLig-1 identified by LC-MS and NMR analysis showed the same as the natural structure of oxLig-1.There were no differences in THP -1 cell apoptosis, as evidenced by FCM at a high concentration of oxLig-1 (60 mg/L).The mRNA and protein expressions of MCP-1 were increased in a dose-dependent manner of oxLig-1, and the maximum value was observed at the concentration of 40 mg/L.ConclusionThe oxLig-1 could upregulate the expression level of MCP-1 in human THP-1 cells.

Abstract:Lipid accumulation and inflammatory reaction play a key role in the process of atherosclerosis, which promote and influence each other. The Adipophilin as cytoplasmic lipid droplets of imperfect-coating protein, not only can be used as a marker of atherosclerotic lipid accumulation, but also can promote lipid accumulation, and inhibit the efflux of intracellular cholesterol, and mediate the secretion of some inflammatory mediators, such as TNF-α, MCP-1 and IL-6, and its expression is regulated by various factors, like the nuclear receptors, fatty acid, modified lipoprotein and hormone-like substance. The purpose of this paper is to make an overview of how Adipophilina plays a role in lipid accumulation and inflammatory response in order to help find a new thought about preventing Atherosclerosis through regulating.

Abstract:Ischemic postconditioning had been observed to reduce infarct size and cell apoptosis on myocardial ischemia-reperfusion injury, and was also associated with a reduction in endothelial cell dysfunction and so on. Postconditioning is a fairly simple strategy, and seems possible to be applied in patients. We will review the advancements of protective effects and the mechanisms in myocardial ischemic postconditioning. The operability of postconditioning in clinical application makes it the focus and hotspots in the field of cardiovascular research in recent years. We will review the clinical application and study progress of myocardial ischemic postconditioning.
