    摘要:

    目的 研究转化生长因子α(TGF-α)对人内皮祖细胞(EPC)分化功能的影响并初步揭示其作用机制。方法 从健康人外周血中分离EPC,分别用含0、1、10 μg/L浓度的TGF-α完全培养基诱导培养。观察各组中EPC分化的形态变化,并利用流式细胞术检测各组EPC培养分化过程中CD34+/CD133+和CD31+/vWF+阳性细胞率,Real-time PCR方法测定各组中EPC分化的内皮细胞特异性基因内皮型一氧化氮合酶(eNOS)和Flk-1/KDR表达差异,并通过测定各组中一氧化氮(NO)含量变化比较EPC分化内皮细胞的功能,Western Blot检测各组EPC分化中TGF-α受体EGFR和血管内皮生长因子(VEGF)的表达。结果 用TGF-α诱导培养分离的人EPC,细胞形态能够更快地由干细胞球形分化成梭形;流式细胞仪检测发现与空白对照组相比,1、10 μg/L TGF-α组中3天时EPC标记CD34+/CD133+阳性率和7天时分化的内皮细胞标记CD31+/vWF+阳性率均显著增高,差异有统计学意义(P<0.05);Real-time PCR检测各TGF-α组EPC分化的内皮细胞特异性基因eNOS和Flk-1/KDR表达量也显著上升(P<0.05);1、10 μg/L TGF-α诱导培养组中EPC分化的内皮细胞分泌NO含量显著升高(P<0.05);1、10 μg/L TGF-α诱导培养能够显著促进EPC分化过程中EGFR和VEGF的表达(P<0.05)。结论 TGF-α能够显著促进人EPC的增殖分化及分化内皮细胞的功能,并通过与受体EGFR结合刺激VEGF表达发挥作用。

    Abstract:

    Aim To investigate the influence of transforming growth factor-α (TGF-α) on the differentiation capacity of human endothelial progenitor cells(EPC) and to explore its mechanism of action. Methods EPC were isolated from peripheral blood of healthy people, and cultured with complete medium containing 0, 1, 10 μg/L concentration of TGF-α. Morphological changes were observed in each group in EPC differentiation. By using the flow cytometry, the CD34+/CD133+ and CD31+/vWF+ positive cells were detected during the differentiation of cultured EPC with TGF-α. The expression of eNOS and Flk-1/KDR genes, the specific genes of EPC differentiated endothelial cells, were determinated by Real-time PCR. The NO content changes in each group were detected. The TGF-α receptor (EGFR) and vascular endothelial growth factor (VEGF) expression in EPC differentiation were tested by Western Blot. Results The cell morphology of EPC induced and cultured by TGF-α, could faster differentiate stem cells from the spherical to spindle cells.
