    摘要:

    为观察不同剂量32 P照射对兔自体移植静脉内膜增生和狭窄的预防作用,用苏木精—伊红染色、增殖细胞核抗原免疫组织化学染色、计算机图像分析和透射电镜等观察自体移植静脉在32 P(每平方厘米2、4、8、1 6 μCi)照射下内膜面积、平滑肌细胞增殖和管腔相对丧失率。结果发现,1 6 μCi照射组无明显新生内膜形成,但细胞大量坏死,血管壁结构破坏严重;8μCi照射组少量新生内膜形成,血管壁结构完整,内膜面积、平滑肌细胞增殖指数和管腔相对丧失率明显低于对照组;4 μCi照射组和2 μCi照射组上述指标与对照组无明显差别。结果提示低剂量32 P照射可有效抑制自体移植静脉内膜增生和狭窄,本模型的适宜剂量为8μCi cm2 。

    Abstract:

    Aim To observe the inhibitory effect of different doses 32 P radiation on the neointimal hyperplasia and stenosis of antologous vein graft. Methods Thirty rabbits underwent 32 P radiotherapy with different doses of 2,4,8,16 μCi/cm 2. HE staining, proliferating cell nuclear antigen immunohistochemical method and computer imaging analysis technique were performed. The neointimal area, the smooth muscle cell (SMC) proliferation index and vessel cavities lost rate were analyzed. Results No neointima was observed in 16 μCi/cm 2 group, but necrosis of most vessel wall cells occurred and the vessels were injured seriously. Thin neointima was formed in 8 μCi/cm 2 group. The neointimal area, SMC proliferation index and cavities lost rate were less than in the control group. There was no significant difference in the parameters between the 2 μCi/cm 2 group, 4 μCi/cm 2 group and control group. Conclusion Low dose 32 P radiation could effectively inhibit the neointimal hyperplasia and restenosis of autologous vein grafts, and the suitable dose was 8 μCi/cm 2 in this model.
